Usually the development process consists of 4 steps:
Step 1: Synthesis of the detection antibody-DNA conjugate
Chimera has developed proprietary technologies for the optimal design and synthesis of antibody-DNA conjugates. The antibody-DNA-conjugate provides the foundation for any given Imperacer® assay format.
Step 2: Conjugate functionality study
The functionality study represents the starting point for an assay optimization. Initial experiments indicate the efficiency of the materials and predict the level of success. This allows for an early milestone to reduce your costs and efforts. The functionality study includes:
- Comparison to an ELISA
- First variation of assay parameters
- Evaluation of starting points for optimization
- Brief data sheet summarizing the results and recommended next steps
Step 3: Assay set up and optimization
The assay optimization comprises all steps needed for an optimal performance of the Imperacer® assay:
- Systematic variation of assay parameters
- Adaptation to biological matrix
- Optimization of signal-to-noise ratio
- A detailed report describing all results
Step 4: Assay validation
The validation process is carried out under GCP/GLP-conform conditions and addresses:
- Intra- and interassay precision
- Sensitivity, detection limit and quantification range
- Accuracy and recovery
- Specificity and selectivity
- Robustness
- GLP/GCP-appropriate SOPs
- A detailed validation report
